Keynote Paper: Cell-Surface Adhesive Interactions in Microchannels and Microvessels

چکیده

Adhesive interactions between white blood cells and the interior surface of the blood vessels they contact is important in inflammation and in the progression of heart disease. Parallel-plate microchannels have been useful in characterizing the strength of these interactions, in conditions that are much simplified over the complex environment these cells experience in the body. Recent computational and experimental work by several laboratories have attempted to bridge this gap between behavior observed in flow chamber experiments, and cellsurface interactions observed in the microvessels of anesthetized animals.

Experimental investigations in development of 3D microchannels through ultrasonic micromachining

The ability to fabricate complex structures and patterns at micron and nano level has facilitated use of many miniature devices in various applications. Fluid mixing is one of the major requirements in such microfluidic devices [1]. Efficient mixing of two and more fluids was made possible by development of 3 D microchannels [2, 3]. Effective mixing was observed due to chaotic advection. Furthe...

Mechanotransduction and flow across the endothelial glycocalyx.

In this inaugural paper, we shall provide an overview of the endothelial surface layer or glycocalyx in several roles: as a transport barrier, as a porous hydrodynamic interface in the motion of red and white cells in microvessels, and as a mechanotransducer of fluid shearing stresses to the actin cortical cytoskeleton of the endothelial cell. These functions will be examined from a new perspec...

Microvasculature on a chip: study of the Endothelial Surface Layer and the flow structure of Red Blood Cells

Microvasculatures-on-a-chip, i.e. in vitro models that mimic important features of microvessel networks, have gained increasing interest in recent years. Such devices have allowed investigating pathophysiological situations involving abnormal biophysical interactions between blood cells and vessel walls. Still, a central question remains regarding the presence, in such biomimetic systems, of th...

The hydrodynamically relevant endothelial cell glycocalyx observed in vivo is absent in vitro.

In recent years, the endothelial cell surface glycocalyx has emerged as a structure of fundamental importance to a broad range of phenomena that determine cardiovascular health and disease. This new understanding of the functional significance of the glycocalyx has been made possible through recently developed experimental techniques using intravital microscopy that are capable of directly prob...
